    〔摘 要〕 目的:研究西达本胺对耐伊马替尼慢性髓系白血病(CML)细胞株 K–562(K–562/Ima)细胞增殖、凋亡 的影响。方法:根据预实验结果,选定西达本胺(作用时间 48 h)不同浓度(终浓度 0、5、10、20、30 μmol·L-1)对 K–562/Ima 细胞进行干预,CCK–8 法检测细胞活力即增殖能力。根据 CCK–8 实验结果,选定西达本胺(终浓度为 0、1、3、 5、7 μmol·L-1),对 K–562/Ima 细胞进行干预,流式细胞术检测细胞的凋亡。结果:西达本胺作用 K–562/Ima 细胞 48 h 后, 不同浓度均可显著抑制 K–562/Ima 细胞的增殖,且诱导 K–562/Ima 细胞凋亡,随着浓度增加,细胞增殖抑制率、凋亡率明 显升高。结论:西达本胺对 K–562/Ima 细胞有显著的抑制增殖、促进凋亡作用,且均有浓度依赖性。

    Abstract:

    〔Abstract〕 Objective The purpose of this study was to observe the effect of chidamide on the proliferation and apoptosis of K–562/Ima cell which is imatinib-resistant chronic myeloid leukemia cell lines. Methods According to the results of the preliminary experiment, the K–562/Ima cells were respectively treated by chidamide with different concentrations (fi nal concentration 0, 5, 10, 20, 30 μmol·L-1) for 48h. The cells proliferation inhibition rates were measured by CCK-8 method. Then, according to the results of the CCK-8 method, the K–562/Ima cells were treated by chidamide with different concentrations (fi nal concentration 0, 1, 3, 5, 7 μmol·L-1) for 48h. The apoptosis rate was detected by fl ow cytometry. Results Different concentrations of chidamide were treated with K–562/Ima cells for 48 hours respectively. It was found that the inhibitory rate of cell proliferation was signifi cantly increased with the increase of chidamide concentration, and the rate of cell apoptosis was apoptosis signifi cantly increased with the increase of chidamide concentration. Conclusion Cedabide can signifi cantly inhibit the proliferation and promote the apoptosis of K–562/Ima cells in a concentration-dependent manner.
